What MTT Actually Measures

MTT is reduced to purple formazan by cellular reductases, mainly NAD(P)H-dependent. The signal therefore reflects metabolic activity, not cell number directly — a distinction that matters more often than it is acknowledged.

Viability % = (Atreated − Ablank) / (Acontrol − Ablank) × 100
SituationEffect on MTT signal
Metabolic inhibition without deathFalsely low viability
Compound stimulates metabolismFalsely high — can exceed 100%
Cell cycle arrest, cells aliveReduced signal, no death
Compound reduces MTT chemicallyFalse signal — ascorbate, some polyphenols

Directly reducing compounds are the most dangerous artefact, because they generate formazan without any cells present. A cell-free control containing compound and MTT detects this and should be routine for any novel chemistry.

Why IC50 Is Not a Constant

IC50 depends on assay duration, seeding density, and readout. A 72-hour exposure gives a lower IC50 than 24 hours for most cytotoxics, and denser cultures shift it upward.

VariableEffect on IC50
Longer exposureLowers
Higher seeding densityRaises
Serum concentrationRaises for protein-bound drugs
Readout methodShifts systematically

IC50 values are therefore not comparable across protocols, and publications should state duration, density and assay. Comparisons within a single experiment are meaningful; comparisons against literature values often are not.

Fitting the Curve

IC50 should be obtained from a four-parameter logistic fit across concentrations spanning the full response, ideally with the top and bottom plateaus defined. Interpolating between two points near 50% assumes a slope the data has not established.

A Hill slope well above 1 suggests cooperative action or a threshold effect; well below 1 suggests a heterogeneous population or multiple targets. The slope carries information and should be reported alongside the IC50.

Worked Examples

Example 1: Control=0.90, treated=0.45, blank=0.05
Viability=(0.45-0.05)/(0.90-0.05)×100
Result: Viability=47% — above IC50
Drug at or above IC50 concentration
Example 2: Blank=0.05, control=0.90, treated=0.92
Viability=102% — slightly above 100%
Result: Within assay variation (±10%)
No cytotoxic effect at this concentration
Example 3: A false positive from chemistry
Antioxidant compound, cell-free wells show colour
Result: Compound reduces MTT directly
The apparent viability signal is chemical, not cellular. A cell-free control reveals it immediately.
Example 4: Why duration changes IC50
Same drug at 24 versus 72 hours
Result: IC50 falls with longer exposure
Cytotoxic effects accumulate. Reporting an IC50 without the exposure time makes it uninterpretable.

Common Mistakes

⚠️
Treating MTT signal as cell number

It measures metabolic reductase activity. Cells arrested but alive give reduced signal without any death occurring.

⚠️
Omitting the cell-free compound control

Ascorbate and various polyphenols reduce MTT directly, generating formazan with no cells present. This control is essential for novel chemistry.

⚠️
Comparing IC50 values across protocols

IC50 depends on exposure duration, seeding density and serum concentration. Literature comparisons require matched conditions.

⚠️
Estimating IC50 from two points

A proper four-parameter fit across the full concentration range is needed. Two-point interpolation assumes a slope the data has not established.

Frequently Asked Questions

MTT vs other viability assays?
MTT (thiazolyl blue): yellow→purple, measures mitochondrial dehydrogenase. Requires DMSO to dissolve formazan. MTS (CellTiter 96): soluble, no dissolution step. CCK-8: WST-8 reagent, more sensitive. CellTiter-Glo: ATP-based, most sensitive, 96/384-well compatible.
IC50 calculation?
Dose-response curve: sigmoid (logistic). IC50 = concentration for 50% inhibition. Use nonlinear regression: 4-parameter logistic (4PL) in GraphPad Prism, R (drc package), or Python (scipy). Run minimum 8 concentrations in triplicate for reliable IC50.
What does the MTT assay measure?
Mitochondrial and cytosolic reductase activity, which correlates with viability but is not the same as cell number.
Why can viability exceed 100%?
Either normal assay variation of about 10%, or a compound that stimulates metabolic activity without increasing cell number.
Why include a cell-free control?
Because some compounds reduce MTT directly, producing formazan with no cells present. Ascorbate and several polyphenols do this.
Are IC50 values comparable between papers?
Often not. IC50 depends on exposure duration, seeding density, serum content and readout, so matched conditions are required for valid comparison.
